"I'm her biggest fan. I love what she brings to this role; there's such a badass in her," Arquette told PEOPLE at Tuesday's Scream 4 premiere in New York.
The pair worked together on the movie, which Arquette said was a very positive experience. "We had a great time," he said. "We're really supportive and loving and kind to each other, so that's really what's important."
Arquette, 39, said he was thrilled to work closely again with Cox, 46, playing the married couple Dewey Riley and Gale Weathers. "I loved acting opposite Courteney," he said. "We don't have that opportunity a lot, and it's a different way of working together. We have so much fun."
His favorite moment? The scene of their first argument. "The second two films, I wasn't in the Dewey cop uniform," Arquette said. "So, to be back in the uniform, having this argument with Courteney, on camera, and seeing her get all feisty, just was really fun."
Although the couple remain separated, Arquette said "everything's great" between them.
He added: "We're still in the same situation that we were before, and nothing's really changed. But we're really being kind and supportive of each other. If anybody's going through a situation like this, that's the way to deal with it. If you bring resentment toward it, or use kids or finances or any negative behavior, it tends to make the whole process a lot more difficult, and it's difficult already ... She's my best friend, so that's really it."
